In the early 90’s MIT post-doctoral researcher Shuguang Zhang observed an unusual repeating peptide sequence while studying the peptide Zuotin, which binds to Z-DNA. This unique repeating sequence led to the discovery of self-assembling peptides. This was an entirely new field for use in medical applications. PuraStat is a synthetic haemostatic material in the form of a prefilled syringe. PuraStat is symbolized for haemostasis in the subsequent situations encountered during surgery when haemostasis by ligation or standard means is inadequate or impractical:
- Bleeding from diminutive blood vessels and oozing from capillaries of the parenchyma of solid organs
- Leaking from vascular anastomoses
- Bleeding from small vessels and seeping from capillaries of the GI tract
- Attenuation of delayed bleeding following gastrointestinal endoscopic submucosal dissection (ESD) procedures in the colon
In the PuraStat® Technology, the matrix is formed from synthetic peptides composed of three naturally occurring amino acids in a repeated sequence four times. The peptides self-assemble to create a 3-dimensional matrix upon a change in pH2 due to the exposure to an ionic solution such as peritoneal fluid, blood, or other bodily secretions.
Whether it is a routine polypectomy or a more complicated ESD procedure, intraprocedural bleeding management, particularly delayed bleeding, remains a significant challenge for endoscopists. If bleeding is not managed efficiently during the procedure, it can lead to delayed bleeding.

Challenges During Endoscopic Procedures include bleeding as the most frequent complication during endoscopic procedures. For intra-procedural bleeds, some solutions such as clips and cauterisation are not fitting for the bleeding location, particularly in small difficult to reach areas. Maintaining clear visibility during haemostasis is essential, especially when working within a tightly enclosed space.
There is also a risk of delayed bleeding after an endoscopic procedure, especially after endoscopic submucosal dissection (ESD). If untreated, delayed bleeding can result in serious conditions such as haemorrhagic shock. The Benefits of PuraStat® include an easy-to-use, transparent hydrogel that enables precise work in a narrow space without compromising the view during endoscopic procedures. It is inert, non-adhesive, and effectively deliverable through an endoscopic applicator. PuraStat can also add value when used in amalgamation with clips and cauterization.
PuraStat has also been proven to decrease the rate of delayed bleeding following colonic ESD by 50%.
For application within gastro-enterology, PuraStat is indicated for:
- Achieving haemostasis in bleeding from small blood vessels and oozing from capillaries of the GI tract following surgical procedures [when haemostasis by ligation or standard means is insufficient or impractical]
- Reduction of delayed bleeding following gastrointestinal endoscopic submucosal dissection (ESD) procedures in the colon.
